CD4抗原是一种55 kDa的细胞表面I型膜糖蛋白,属于免疫球蛋白超家族。CD4作为协同受体,与T细胞受体(TCR)协同作用,与抗原呈递细胞(APC)展示的ⅱ类MHC分子相互作用。CD4由大多数胸腺细胞和辅助性T细胞(NK-T细胞的一个亚群)表达,并且由树突细胞和巨噬细胞微弱地表达。CD4在T细胞的发育中起着重要作用,是成熟T细胞发挥最佳功能所必需的。据报道,RPA-T4抗体结合CD4的D1结构域,并且不阻断OKT-4抗体的结合。此外,RPA-T4已被证明能阻断HIV gp120蛋白与CD4的结合,并在体外抑制CD4 T细胞的活化。
