
3,3′-Diaminobenzidine (DAB) is an organic compound with the formula (C6H3(NH2)2)2. This derivative of benzidine is a precursor to polybenzimidazole, which forms fibers that are renowned for their chemical and thermal stability. Diaminobenzidine is oxidized by hydrogen peroxide in the presence of hemoglobin to give a dark-brown color.

Store at 2−8 °C and protected from light. Do not freeze. The expiration date is indicated on the vial label.

Protocol:
Add on drop or 50ul DAB chromogen into 1ml DAB substrate buffer. Use within 2 hours after preparation.
Apply enough of DAB working solution to cover the specimen completely. Incubate 5-10 minutes. Monitor the color development under light microscope. Rinse slides gently with distilled water.
