Flow cytometric analysis of CD33 expression on human peripheral blood leukocytes. Whole blood was stained with either BD Horizon™ BV421 Mouse Anti-Human CD33 antibody (Cat. No. 562854/565949; solid line histogram) or with a BD Horizon™ BV421 Mouse IgG1, k Isotype Control (Cat. No. 562438; dashed line histogram). The erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). The fluorescence histograms were derived from monocytes (Left Panel) or granulocytes (Right Panel) with the light scattering properties of viable cells. Flow cytometry was performed using a BD™ LSR II Flow Cytometer System.
Multiparameter flow cytometric analysis of CD33 expression on human peripheral blood leucocyte populations. Human whole blood was stained with either BD Horizon™ BV421 Mouse IgG1, κ Isotype Control (Cat. No. 562438; Left Plot) or BD Horizon™ BV421 Mouse Anti-Human CD33 antibody (Cat. No. 565941/562854; Right Plot). The erythrocytes were lysed with BD FACS™ Lysing Solution (Cat. No. 349202). A bivariate pseudocolor density plot showing the correlated expression of CD33 (or Ig Isotype control staining) versus side light-scatter (SSC) signals was derived from gated events with the forward and side light-scatter characteristics of intact leucocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ Flow Cytometer System and FlowJo™ software.
